When are yellow roadwork signs mandatory?

They are mandatory every time a job, a deposit of materials or a machine occupies part of the roadway or alters its viability, even for a few hours.

Do construction site signs have to be reflective?

Yes, absolutely. Temporary signs must also be reflective (Class 1 or Class 2) to be visible at night, just like permanent signs.

Do I need to cover the existing white signs?

Yes. Art. 21 provides that permanent signs in contrast to temporary ones (e.g. an old limit of 50 km/h when the construction site imposes 30 km/h) must be obscured or removed so as not to create confusion for drivers.
